Real-time monitoring of brake shoe keys in freight cars

摘要: Condition monitoring ensures the safety of freight railroad operations. With the development of machine vision technology, visual inspection has become a principal means of condition monitoring. The brake shoe key (BSK) is an important component in the brake system, and its absence will lead to serious accidents. This paper presents a novel method for automated visual inspection of the BSK condition in freight cars. BSK images are first acquired by hardware devices. The subsequent inspection process is divided into three stages: first, the region-of-interest (ROI) is segmented from the source image by an improved spatial pyramid matching scheme based on multi-scale census transform (MSCT). To localize the BSK in the ROI, census transform (CT) on gradient images is developed in the second stage. Then gradient encoding histogram (GEH) features and linear support vector machines (SVMs) are used to generate a BSK localization classifier. In the last stage, a condition classifier is trained by SVM, but the features are extracted from gray images. Finally, the ROI, BSK localization, and condition classifiers are cascaded to realize a completely automated inspection system. Experimental results show that the system achieves a correct inspection rate of 99.2% and a speed of 5 frames/s, which represents a good real-time performance and high recognition accuracy.

Imbalanced fault diagnosis of rotating machinery using autoencoder-based SuperGraph feature learning

摘要: Existing fault diagnosis methods usually assume that there are balanced training data for every machine health state. However, the collection of fault signals is very difficult and expensive, resulting in the problem of imbalanced training dataset. It will degrade the performance of fault diagnosis methods significantly. To address this problem, an imbalanced fault diagnosis of rotating machinery using autoencoder-based SuperGraph feature learning is proposed in this paper. Unsupervised autoencoder is firstly used to compress every monitoring signal into a low-dimensional vector as the node attribute in the SuperGraph. And the edge connections in the graph depend on the relationship between signals. On the basis, graph convolution is performed on the constructed SuperGraph to achieve imbalanced training dataset fault diagnosis for rotating machinery. Comprehensive experiments are conducted on a benchmarking publicized dataset and a practical experimental platform, and the results show that the proposed method can effectively achieve rotating machinery fault diagnosis towards imbalanced training dataset through graph feature learning.

Dynamic simulation of gas turbines via feature similarity-based transfer learning

摘要: Since gas turbine plays a key role in electricity power generating, the requirements on the safety and reliability of this classical thermal system are becoming gradually strict. With a large amount of renewable energy being integrated into the power grid, the request of deep peak load regulation for satisfying the varying demand of users and maintaining the stability of the whole power grid leads to more unstable working conditions of gas turbines. The startup, shutdown, and load fluctuation are dominating the operating condition of gas turbines. Hence simulating and analyzing the dynamic behavior of the engines under such instable working conditions are important in improving their design, operation, and maintenance. However, conventional dynamic simulation methods based on the physic differential equations is unable to tackle the uncertainty and noise when faced with variant real-world operations. Although data-driven simulating methods, to some extent, can mitigate the problem, it is impossible to perform simulations with insufficient data. To tackle the issue, a novel transfer learning framework is proposed to transfer the knowledge from the physics equation domain to the real-world application domain to compensate for the lack of data. A strong dynamic operating data set with steep slope signals is created based on physics equations and then a feature similarity-based learning model with an encoder and a decoder is built and trained to achieve feature adaptive knowledge transferring. The simulation accuracy is significantly increased by 24.6% and the predicting error reduced by 63.6% compared with the baseline model. Moreover, compared with the other classical transfer learning modes, the method proposed has the best simulating performance on field testing data set. Furthermore, the effect study on the hyper parameters indicates that the method proposed is able to adaptively balance the weight of learning knowledge from the physical theory domain or from the real-world operation domain.

Build orientation determination of multi-feature mechanical parts in selective laser melting via multi-objective

摘要: Selective laser melting (SLM) is a unique additive manufacturing (AM) category that can be used to manufacture mechanical parts. It has been widely used in aerospace and automotive using metal or alloy powder. The build orientation is crucial in AM because it affects the as-built part, including its part accuracy, surface roughness, support structure, and build time and cost. A mechanical part is usually composed of multiple surface features. The surface features carry the production and design knowledge, which can be utilized in SLM fabrication. This study proposes a method to determine the build orientation of multi-feature mechanical parts (MFMPs) in SLM. First, the surface features of an MFMP are recognized and grouped for formulating the particular optimization objectives. Second, the estimation models of involved optimization objectives are established, and a set of alternative build orientations (ABOs) is further obtained by many-objective optimization. Lastly, a multi-objective decision making method integrated by the technique for order of preference by similarity to the ideal solution and cosine similarity measure is presented to select an optimal build orientation from those ABOs. The weights of the feature groups and considered objectives are achieved by a fuzzy analytical hierarchy process. Two case studies are reported to validate the proposed method with numerical results, and the effectiveness comparison is presented. Physical manufacturing is conducted to prove the performance of the proposed method. The measured average sampling surface roughness of the most crucial feature of the bracket in the original orientation and the orientations obtained by the weighted sum model and the proposed method are 15.82, 10.84, and 10.62 μm, respectively. The numerical and physical validation results demonstrate that the proposed method is desirable to determine the build orientations of MFMPs with competitive results in SLM.

提升KPCA方法特征抽取效率的算法设计

摘要:

在PCA基础上发展出的KPCA方法能抽取样本的非线性特征分量。然而, 基于KPCA的特征抽取需计算所有训练样本与待抽取特征的样本间的核函数, 因此, 训练集的大小制约着特征抽取的效率。为了提高效率,假设特征空间中变换轴可由一部分训练样本(节点)线性表出,并设计了改进的KPCA算法(IKPCA)。该算法抽取某样本特征时,只需计算该样本与节点间的核函数即可。实验结果显示,IKPCA在对应较好性能的同时,具有明显的效率上的优势。

composition differences between processed protein from different animal species by self-organizing feature

摘要: Amino acids are the dominant organic components of processed animal proteins, however there has been limited investigation of differences in their composition between various protein sources. Information on these differences will not only be helpful for their further utilization but also provide fundamental information for developing species-specific identification methods. In this study, self-organizing feature maps (SOFM) were used to visualize amino acid composition of fish meal, and meat and bone meal (MBM) produced from poultry, ruminants and swine. SOFM display the similarities and differences in amino acid composition between protein sources and effectively improve data transparency. Amino acid composition was shown to be useful for distinguishing fish meal from MBM due to their large concentration differences between glycine, lysine and proline. However, the amino acid composition of the three MBMs was quite similar. The SOFM results were consistent with those obtained by analysis of variance and principal component analysis but more straightforward. SOFM was shown to have a robust sample linkage capacity and to be able to act as a powerful means to link different sample for further data mining.

Blockage of receptor-interacting protein 2 expression by small interfering RNA in murine macrophages

摘要: This study aims to demonstrate that blocking the receptor-interacting protein2 (Rip2) expression can decrease inflammatory cytokine production by macrophage and protect mice from endotoxin lethality. Murine Rip2 small interfering RNA (siRNA) plasmids were constructed and transfected into macrophage and Rip2 expression was detected with reverse transcription-polymerase chain reaction (RT-PCR) and western blot. Cell proliferation was assayed with MTT. TNF-? concentration was assayed with ELISA and high-mobility group box 1 protein (HMGB1) level with semi-quantitative western blot after lipopolysaccharide (LPS) stimulation. LPS challenge was given after the plasmids were injected into mice and the survival rate was calculated. Rip2 siRNA plasmid could block the mRNA and protein expression of Rip2 and promote cell proliferation. Blocking Rip2 could attenuate LPS-induced TNF-? and HMGB1 production. The HMGB1 expression in the liver decreased to (40.21 ± 11.03) pg/g, and serum TNF-? level decreased to (300.43 ± 59.26) ng/L ( < 0.05). The survival rate of mice from endotoxemia was also improved ( < 0.05). The results demonstrate that Rip2 siRNA plasmid can block the expression of Rip2, decrease the production of TNF-? and HMGB1 and protect mice from fatal endotoxemia.

MSI/LOH and extron expression of the FHIT gene in gastric carcinoma

摘要: We detected loss of heterozygosity (LOH) and microsatellite instabilities (MSI), as well as extron expression of the fragile histidine triad (FHIT) gene in gastric carcinoma (GC), in order to evaluate their association with clinicopathological processes in gastric carcinogenesis. LOH and MSI of the FHIT were detected by using PCR at 4 microsatellite loci: D3S 1300, D3S 4103, D3S 1481, D3S 1234 in cancer tissues from 50 patients with primary GC, with normal mucosa acting as matched controls. FHIT transcripts were detected by nested RT-PCR in 30 cases of GC and their products were sequenced. Results show that the average frequencies of LOH and MSI of the FHIT gene in GC were 32.4% and 26.4%, respectively. There was no correlation between LOH and MSI of the FHIT gene in GC and the histological characteristics of gastric carcinoma (Bormann s or Lauren s classification). LOH of the FHIT gene in GC was related to depth invasiveness, and its frequency in GC where serosa was penetrated was significantly higher than that in GC without serosa penetration (73.5% 37.5%, <0.05). The frequency of MSI in GC without lymph node metastasis was significantly higher than that in GC with lymph node metastasis (66.7% 34.3%, <0.05). Aberrant transcripts were found in 11/30 GC tissues. Sequencing analysis of the aberrant fragments found a RT-PCR product missing exons 5 7 in one case of GC, and another product missing exons 4 7. Four of 10 (40.0%) cases of primary GC showed absent or decreased expression of the FHIT protein as compared to their matched normal tissues. The findings in this study suggest that LOH and MSI of FHIT gene may induce aberrant extron expression, which might play a role in gastric carcinogenesis.

Gene expression disparity in giant cell tumor of bone

摘要: The aim of this paper was to study the differential gene expression of giant cell tumor of bone (GCTB) by gene chip technology. Total RNA of 8 fresh GCTB specimens (Jaffe I∶6 cases, II∶1 case, III∶1 case; Campanacci I∶6 cases, II∶1 case, III∶1 case; Enneking Staging G T M : 5 cases, G T M : 2 cases, G T M : 1 case) and 4 normal bony callus specimens (the control group) were extracted and purified to get mRNA and then reverse transcribed to complementary DNA, respectively. Microarray screening with a set of 8064 human cDNA genes was conducted to analyze the difference among the samples and the control. The hybridization signals were scanned. The gene expression disparity between the GCTB samples and normal bony callus was significantly different ( <0.01), and the disparity of over 5-fold was found in 47 genes in the GCTB specimens, with 25 genes up-regulated and 22 down-regulated including the extracellular matrix and transforming-related genes, oncogene and its homolog genes, cytokine and its receptor genes. Specific gene spectrum associated with GCTB can be identified by cDNA microarray, which will be the foundation of progressive etiology elucidation, diagnosis and treatment of GCTB.

Distinct gene expression pattern of mutations coordinated by target repression and promoter hypermethylation

摘要: Runt-related transcription factor 1 (RUNX1) is an essential regulator of normal hematopoiesis. Its dysfunction, caused by either fusions or mutations, is frequently reported in acute myeloid leukemia (AML). However, RUNX1 mutations have been largely under-explored compared with RUNX1 fusions mainly due to their elusive genetic characteristics. Here, based on 1741 patients with AML, we report a unique expression pattern associated with RUNX1 mutations in AML. This expression pattern was coordinated by target repression and promoter hypermethylation. We first reanalyzed a joint AML cohort that consisted of three public cohorts and found that RUNX1 mutations were mainly distributed in the Runt domain and almost mutually exclusive with NPM1 mutations. Then, based on RNA-seq data from The Cancer Genome Atlas AML cohort, we developed a 300-gene signature that significantly distinguished the patients with RUNX1 mutations from those with other AML subtypes. Furthermore, we explored the mechanisms underlying this signature from the transcriptional and epigenetic levels. Using chromatin immunoprecipitation sequencing data, we found that RUNX1 target genes tended to be repressed in patients with RUNX1 mutations. Through the integration of DNA methylation array data, we illustrated that hypermethylation on the promoter regions of RUNX1-regulated genes also contributed to dysregulation in RUNX1-mutated AML. This study revealed the distinct gene expression pattern of RUNX1 mutations and the underlying mechanisms in AML development.

Immunohistochemical measurement and expression of Mcl-1 in infertile testes

The diagnosis of azoospermia represents a major challenge to andrologists as this condition may occur despite normal spermatogenesis and genital tracts. Myeloid cell leukemia-1 (Mcl-1) is a member of the Bcl-2 family of proteins involved in regulation of apoptosis in various cell types. This study aimed to investigate the immunohistochemical expression of Mcl-1 in testicular biopsies of subjects with azoospermia. Eighty-six cases with azoospermia were obtained from 509 infertile patients admitted to the Andrology Unit of the Zagazig University Hospitals from January 2010 to December 2011. Biopsies were diagnosed and classified using H&E-stained slide sections. The specimens were subjected to immunohistochemical staining for Mcl-1 and examined through light microscopy. Forty-five cases of maturation arrest (25 at spermatids and 20 at the spermatocytes), 31 cases of hypospermatogenesis (20 moderate and 11 severe), 5 cases of Sertoli?cell-only syndrome, 2 cases of basement membrane hyalinization, and 1 case of tubular and peritubular sclerosis were observed. Normal spermatogenesis was detected in 2 cases. A strong positive immunoreaction in Leydig cells was observed among all investigated specimens. A moderate reaction was detected in spermatocytes and spermatozoa in cases of normal spermatogenesis and hypospermatogenesis, but a negative reaction was detected in cases of maturation arrest and germ cell aplasia. Apoptosis was found to be associated with decreased rate of spermatogenesis. High apoptosis rates may result in azoospermia, which can occur despite normal spermatogenesis and absence of duct obstruction.

Construction of a universal recombinant expression vector that regulates the expression of human lysozyme

The mammary gland provides a novel method for producing recombinant proteins in milk of transgenic animals. A key component in the technology is the construction of an efficient milk expression vector. Here, we established a simple method to construct a milk expression vector, by a combination of homologous recombination and digestion-ligation. Our methodology is expected to have the advantages of both plasmid and bacterial artificial chromosome (BAC) vectors. The BAC of mouse whey acidic protein gene (mWAP) was modified twice by homologous recombination to produce a universal expression vector, and the human lysozyme gene (hLZ) was then inserted into the vector by a digestion-ligation method. The final vector containing the 8.5 kb mWAP 5′ promoter, 4.8 kb hLZ genomic DNA, and 8.0 kb mWAP 3′ genomic DNA was microinjected into pronuclei of fertilized mouse embryos, to successfully generate two transgenic mouse lines that expressed recombinant human lysozyme (rhLZ) in milk. The highest expression level of rhLZ was 0.45 g·L1, and rhLZ exhibited the same antibacterial activity as native hLZ. Our results have provided a simple approach to construct a universal milk expression vector, and demonstrated that the resulting vector regulates the expression of hLZ in milk.

Relationship of adrenomedullin expression and microvessel density and prognosis in smooth muscle tumor

摘要: The aim of this paper was to investigate the relationship between the expression of adrenomedullin (ADM) and microvessel density (MVD) and prognosis in smooth muscle tumor of uterus. The expression of ADM was detected using immunohistochemical staining in specimens from 15 normal controls, 28 cases of uterine leiomyoma (LE) and 19 cases of uterine leiomyosarcoma (LES). The MVD was assayed by immunostainting with CD. There was a positive correlation between the ADM expression and MVD in LE and LES respectively ( = 0.823, <0.01; = 0.793, <0.01). The expression of ADM in LE was statistically lower than that in LES (<0.05). There was a positive correlation between the ADM expression and mitotic figures in LES (<0.05): the more mitotic figures, the higher levels of the ADM expression and poor prognosis. The ADM is an important angiogenic factor in smooth muscle tumor of uterus. The ADM can be used as an accessory marker in estimating the malignant potency of LE and judging the pro gnosis of LES, and as a novel molecular target of anti-angiogenic and anticarcinogenic strategies.

Fault feature extraction of planet gear in wind turbine gearbox based on spectral kurtosis and time wavelet

Planetary transmission plays a vital role in wind turbine drivetrains, and its fault diagnosis has been an important and challenging issue. Owing to the complicated and coupled vibration source, time-variant vibration transfer path, and heavy background noise masking effect, the vibration signal of planet gear in wind turbine gearboxes exhibits several unique characteristics: Complex frequency components, low signal-to-noise ratio, and weak fault feature. In this sense, the periodic impulsive components induced by a localized defect are hard to extract, and the fault detection of planet gear in wind turbines remains to be a challenging research work. Aiming to extract the fault feature of planet gear effectively, we propose a novel feature extraction method based on spectral kurtosis and time wavelet energy spectrum (SK-TWES) in the paper. Firstly, the spectral kurtosis (SK) and kurtogram of raw vibration signals are computed and exploited to select the optimal filtering parameter for the subsequent band-pass filtering. Then, the band-pass filtering is applied to extrude periodic transient impulses using the optimal frequency band in which the corresponding SK value is maximal. Finally, the time wavelet energy spectrum analysis is performed on the filtered signal, selecting Morlet wavelet as the mother wavelet which possesses a high similarity to the impulsive components. The experimental signals collected from the wind turbine gearbox test rig demonstrate that the proposed method is effective at the feature extraction and fault diagnosis for the planet gear with a localized defect.

Speech emotion recognitionwith unsupervised feature learning

摘要: Emotion-based features are critical for achieving high performance in a speech emotion recognition (SER) system. In general, it is difficult to develop these features due to the ambiguity of the ground-truth. In this paper, we apply several unsupervised feature learning algorithms (including -means clustering, the sparse auto-encoder, and sparse restricted Boltzmann machines), which have promise for learning task-related features by using unlabeled data, to speech emotion recognition. We then evaluate the performance of the proposed approach and present a detailed analysis of the effect of two important factors in the model setup, the content window size and the number of hidden layer nodes. Experimental results show that larger content windows and more hidden nodes contribute to higher performance. We also show that the two-layer network cannot explicitly improve performance compared to a single-layer network.
